TA, Blunt, or Simple?
- Match the polymerase — TA kits (CYCV14, CYCV15) for Taq A-overhang fragments. Blunt kits (CYCV16, CYCV17) for Pfu/KOD/Phusion products.
- Standard vs Simple — Standard vectors (CYCV14, CYCV16) have common restriction sites flanking the cloning site so you can cut the insert out. Simple vectors (CYCV15, CYCV17) omit those flanking sites to avoid redundant restriction sites in the insert.
- Do not 5′-phosphorylate the DNA — all four kits require free 5′-OH ends. Competent cells are not in the box (vector, 1000 bp control, enhancer only).
- Shipping — ice packs; store at −20°C (12 months in the manuals).
Frequently asked questions
- TA or Blunt? If the amplicon came from Taq (including FastLong CYPC80), use CYCV14 or CYCV15. If it came from a high-fidelity blunt polymerase (FastHiFi CYPC82, LongHiFi CYPC84), use CYCV16 or CYCV17.
- What does Simple mean? The Simple vectors do not carry common restriction sites next to the cloning site. Use them when those sites would collide with sites inside your insert.
- Do the kits include competent cells? No. Listed contents are the pTOPO vector, a 1000 bp control insert, and enhancer. Ask if you also need cells.
- How are kits shipped? Ice packs for every CYCV catalog. Store at −20°C on arrival.
- Are these kits for clinical use? No. Research Use Only (RUO). Not for diagnostic, therapeutic, or clinical use.
