How fast is FastHiFi compared with a standard Pfu mix?
FastHiFi amplifies at 2–4 kb/min, up to 4–8× the speed of Pfu, with fidelity up to 54× of traditional Taq and yield up to 1.5–2× of traditional Pfu (manual).
What template lengths can FastHiFi handle?
Per the manual: not more than ~3 kb on complex genomic DNA, and not more than ~6 kb on simple genomes, plasmids or phage DNA. Recommended 50-µl inputs: plasmid 0.1–10 ng, bacterial genome 10–100 ng, human genome 50–150 ng, or 1–5 µl cDNA from an RT reaction.
Are FastHiFi products blunt-ended?
Yes. Products are blunt-ended — use a blunt-end cloning workflow such as our pTOPO Blunt kits, not TA cloning. After purification, products also work for digestion, ligation or sequencing.
How do I amplify high-GC or complex templates?
Raise pre-denaturation and denaturation to 98°C (CYA8 is heat-resistant and its activity is not influenced at 98°C). If amplification is still poor, add DMSO to 1–8% final (step up in 1% increments) or betaine to 1.0–1.7 M final, together with touchdown PCR.
What cycling conditions does the manual propose?
95°C 3 min → 95°C 10 s / 55°C 10–15 s / 72°C 15–30 s per kb × 30 cycles → final extension 72°C 2–5 min. Choose 30–45 s/kb on complex genomes such as human.
