LongHiFi FAQ
When should I choose LongHiFi instead of FastHiFi?
Both are high-fidelity mixes — pick by target length. FastHiFi (CYPC82) is the faster choice for non-long targets (≤3 kb on complex gDNA). Choose LongHiFi when length matters: the manual supports less than ~10 kb on complex genomic DNA and less than ~15 kb on simple templates.
How fast is LongHiFi?
3–4 kb/min. The manual recommends 10–15 s/kb on simple or plasmid templates and 20–25 s/kb on complicated templates. If smearing appears above and below the main band on the gel, the extension time is too long — decrease it.
Can LongHiFi handle GC-rich targets?
Yes — it is suitable for difficult, GC-rich and long templates. Raise pre-denaturation and denaturation to 98°C (no clear influence on CYA9 activity), or add DMSO 1–8% / betaine 1.0–1.7 M with touchdown PCR.
Are LongHiFi products blunt-ended?
Yes — blunt ends for high-fidelity cloning, and the mix contains a red tracer dye for direct gel loading. For long targets where maximum speed matters more than fidelity, see FastLong (CYPC80).
