Input ranges at a glance
| Gel slice | Up to 400 mg agarose per column (TAE or TBE, standard or low-melting); split larger slices |
| Dissolving buffer | 3 volumes Buffer DD per gel volume (100 mg ≈ 100 µl); 6 volumes for >2% agarose gels |
| PCR cleanup mode | 5 volumes Buffer DD per 1 volume PCR sample (same column workflow) |
| Fragment range / capacity | 100 bp to 10 kb; up to 10 µg PCR product per column |
| Isopropanol step | 150 µl per 100 mg gel — improves yield for fragments <500 bp and >4 kb |
| Elution | 50 µl standard, minimum 30 µl; 70–90°C pre-warmed elution buffer raises yield |
| Storage / pack sizes | All components RT, stable 12 months; CYDR0101 (50 preps) · CYDR0102 (100 preps) |
Limits and handling
- For laboratory research use only. Not for diagnostic or therapeutic use.
- Shipped at ambient temperature (no ice packs).
- Intended for DNA of about 100 bp to 10 kb from standard or low-melt agarose in TAE or TBE. Up to 400 mg agarose per spin column.
- Store at room temperature (manual: 12 months). Add ethanol to Buffer WB before first use.
- Residual ethanol on the membrane interferes with ligation and other downstream steps — complete the dry spin.

Gel Extraction Kit FAQ
When does the isopropanol step actually help?
The manual states isopropanol improves yield for fragments smaller than 500 bp and larger than 4 kb; for 500 bp–4 kb fragments it has no effect on yield. Add it after the gel has completely dissolved and do not centrifuge at that stage.
Can I also purify PCR products with this kit?
Yes — the manual includes a PCR cleanup protocol: mix 5 volumes of Buffer DD with 1 volume of PCR sample and follow the same column steps. For routine PCR cleanup you may also want the dedicated PCR Purification Kit (CYDR02).
What agarose concentrations are supported?
TAE or TBE gels, standard or low-melting point. The colored Buffer DD helps you visually confirm dissolving, binding and washing. For >2% gels use 6 volumes of Buffer DD and extend the 56°C incubation until the gel is completely dissolved.
