Typical Applications

  • Total RNA from animal cells or tissues using a gDNA-elimination column (no DNase step in the standard protocol).
  • Manual tissue examples include liver and brain; cultured cells as listed in the protocol.
  • RNA for RT-qPCR where residual genomic DNA is a problem.

Input ranges at a glance

Cultured cells<5×10⁶ cells in 350 μl lysis buffer; 5×10⁶–1×10⁷ cells in 600 μl
Animal tissue<20 mg in 350 μl; 20–30 mg in 600 μl
Manual tissue examplesMouse liver and brain
gDNA removalgDNA elimination column: gDNA binds to the membrane, RNA stays in the flow-through — no DNase step
ProtocolPhenol/chloroform-free; lysis buffer immediately inactivates RNase and DNase
StorageAll components at room temperature; stable 9 months at the indicated temperatures
Pack sizeCYRN2802, 50 preps

All values from the CYRN28 manual (PDF).

Limits and handling

  • For laboratory research use only. Not for diagnostic or therapeutic use.
  • Shipped at ambient temperature (no ice packs).
  • Store at room temperature (manual: 9 months). Add the indicated ethanol before first use.
  • gDNA is removed on the elimination column — a separate DNase digest is usually unnecessary, but follow the manual if your assay is DNA-sensitive.

CYRN28 FAQ

  • Does CYRN28 work on fibrous or dense tissue? The manual documents animal tissues (examples: mouse liver and brain) and cultured cells. It does not claim validation on fibrous or collagen-rich tissue — validate on your tissue type before committing samples.
  • Is a DNase digestion step needed? No. The gDNA elimination column binds genomic DNA while RNA stays in the flow-through, so the standard protocol has no DNase step. For DNA-sensitive assays, follow the manual.
  • Is the protocol phenol/chloroform-free? Yes — the whole process is phenol/chloroform-free, and the lysis buffer immediately inactivates RNase and DNase.
  • How can I improve RNA yield? Preheat the RNase-free water to 70–90°C for the elution step (manual). For elution volumes above 700 μl, load in two passes.
