- Does CYRN28 work on fibrous or dense tissue? The manual documents animal tissues (examples: mouse liver and brain) and cultured cells. It does not claim validation on fibrous or collagen-rich tissue — validate on your tissue type before committing samples.
- Is a DNase digestion step needed? No. The gDNA elimination column binds genomic DNA while RNA stays in the flow-through, so the standard protocol has no DNase step. For DNA-sensitive assays, follow the manual.
- Is the protocol phenol/chloroform-free? Yes — the whole process is phenol/chloroform-free, and the lysis buffer immediately inactivates RNase and DNase.
- How can I improve RNA yield? Preheat the RNase-free water to 70–90°C for the elution step (manual). For elution volumes above 700 μl, load in two passes.
